Its formation occurs extracellularly through the collaboration of a proteic transient framework (the enamel organic matrix), which controls hydroxyapatite crystal growth, morphology and orientation. This matrix is deposited with a small amount of mineral during the secretory stage of amelogenesis. The organic components begin to be degraded in the transition stage and are extensively corrupted, and almost entirely replaced by the inorganic crystallites during maturation stage. The present paper reviews current knowledge on the structural biology of the enamel organic matrix.

Protein-to-protein interactions: criteria defining the assembly of the enamel organic matrix.

Enamel crystallites form in a protein matrix located proximal to the ameloblast cell layer. This unique organic extracellular matrix is constructed from structural protein components biosynthesized and secreted by ameloblasts. To date, three distinct classes of enamel matrix proteins have been cloned. These are the amelogenins, tuftelin, and ameloblastin, with recent data implicating ameloblast...

The amino acid composition of the organic matrix of decalcified fetal bovine dental enamel.

The organic matrix of dental enamel is produced by morphologically characteristic cells of the enamel organ (ameloblasts), which are derived from the stratified epithelial cells of the primitive oral cavity (1). On the basis of its ectodermal origin, as well as its chemical composition, physical properties, and histochemical staining characteristics, the protein component of the organic matrix,...

Electron Microscopic Studies on the Organic Matrix of Enamel and Dentin

In previously published electron microscopic studies on the organic substance of enamel, the organic matrix was depicted in thin sections as a meshwork of delicate fibrils.1'"i " If one compares these structures with photographs obtained by the replica method using mineralized enamel, the original relationship between the organic and the inorganic components of the enamel is difficult to recons...

Posttranslational Amelogenin Processing and Changes in Matrix Assembly during Enamel Development

The extracellular tooth enamel matrix is a unique, protein-rich environment that provides the structural basis for the growth of long and parallel oriented enamel crystals. Here we have conducted a series of in vivo and in vitro studies to characterize the changes in matrix shape and organization that take place during the transition from ameloblast intravesicular matrices to extracellular subu...

Helium ion microscopy of enamel crystallites and extracellular tooth enamel matrix

An unresolved problem in tooth enamel studies has been to analyze simultaneously and with sufficient spatial resolution both mineral and organic phases in their three dimensional (3D) organization in a given specimen. This study aims to address this need using high-resolution imaging to analyze the 3D structural organization of the enamel matrix, especially amelogenin, in relation to forming en...
